使用方法
●使用剂量:小鼠的剂量为0.2ml/10克体重,大鼠的注射剂量为10ml/Kg,腹腔注射。不同品系、不同年龄注射剂量略有差异,请提前做预实验确定麻醉剂量。
●麻醉时间:动物5分钟内可麻醉,维持麻醉约10-40分钟,如手术时间需要延长,可在小鼠苏醒前腹腔补注射0.3ml-0.5ml麻醉剂。

Abstract:
Animalmodels,particularlymice,areusedextensivelytoinvestigateneurologicaldiseases.Basicresearchregardinganimalmodelsofhumanneurologicaldiseaserequiresthattheanimalsexhibithallmarkcharacteristicsofthedisease.Theseincludediseasespecificanatomical,metabolicandbehavioralchanges.Nerveconductionvelocity(NCV)isthepredominantmethodusedtoassessperipheralnervehealth.Normativedataadjustedforage,genderandheightisavailableforhumanpatients;however,thesedataarenotavailableformostrodentsincludingmice.NCVmaybeaffectedbyanimalageandsize,bodytemperature,stimulusstrengthandanesthesia.Whiletheeffectsoftemperature,ageandsizearedocumented,thedirectandindirecteffectsofanesthesiaonNCVarenotwellreported.Ourlaboratoryisprimarilyconcernedwithanimalmodelsofdiabeticneuropathy(DN)andusesNCVtoconfirmthepresenceofneu-ropathy.ToensurethatsubtlechangesinNCVarereliablyassayedandnotdirectlyorindirectlyaffectedbyanesthesia,wecomparedtheeffectsof4commonlyusedanesthetics,isoflurane,ketamine/xylazine,sodiumpentobarbitaland2-2-2tribromoethanolonNCVinacommonlyusedrodentmodel,theC57Bl6/Jmouse.Ourresultsindicatethatoftheanestheticstested,isofluranehasminimalimpactonNCVandisthesafest,mosteffectivemethodofanesthesia.Ourdatastronglysuggestthatisofluraneshouldbecome.
