ThefirstpublicationaboutCationiclipidstotransfectDNAintocellswaspublishedbyPhilipFelgner.Currently,Cationiclipidshavebecomeoneoffewmajorclassofmoleculesinlipidnanoparticle(LNP)drugdeliveryvehicles,mRNAvaccineencapsulation.Cationiclipidsneutralizenegativechargesandfacilitateencapsulationandcelluptakeinlipidnanoparticles(LNP).Usuallyunsaturated,short(<30monomer)hydrocarbonchainsofthelipidsareassociatedwiththehighesttransductionefficiencies.Inaqueousenvironments,theselipidsformmicelleswithpositivelychargedsurfacesthatcomplexwithDNA.TheselipoplexesacttoneutralizechargerepulsionbetweenthecellmembraneandDNAandpromoteendocytosis,similartothemechanismofcationicpolymers.
